Product Description
Ninhydrin, from this perspective, is a triketohydrindene hydrate formed from a diketone structure with two imine groups and a carboxyl group. It is, in general, a versatile reagent known for its characteristic reaction with amino acids, proteins, and primary and secondary amines. The chemical turns deep blue or purple in colour in reaction with the amino group in the amino acids; this reaction is known as Ruhemann’s purple. The ability of ninhydrin to form colored complexes with amino acids and primary amines has secured a place for the compound amongst the most important tools of analytical chemistry, forensic science, and biochemical research. Indeed, no substitute has been found for the methods where detection and quantification of nitrogen-containing compounds are required, since it provides sensitivity, selectivity, and easiness.
Applications
- Amino Acid Analysis: Ninhydrin is used with many chromatography techniques, such as paper chromatography and thin-layer chromatography, to separate and identify the amino acids in samples. It enables detection and quantitation of amino acids due to their characteristic color reactions.
- Forensic Analysis: Ninhydrin is utilized in forensic science in detecting and visualizing latent fingerprints left on porous surfaces like paper and cardboard. Amino acids in the residues of sweat left behind on the surfaces react with ninhydrin to form a chromophore that helps identify fingerprints.
- Protein Quantification: Ninhydrin-based assays are used in biochemical and biomedical studies for the quantification of protein content by detecting the presence of amino acids.
- Peptide Synthesis: Ninhydrin is used to check the coupling reactions during peptide synthesis. It will show whether unreacted amino groups left in peptides or in derivatives of amino acids exist, thus letting one optimize the synthesis.
- Analytical Chemistry: Ninhydrin represents a very sensitive and rather selective reagent for the determination and quantitative estimation of primary amines and similar compounds involved in different analytical techniques.
- Colorimetric Detection: Ninhydrin reaction with amino acids is both very sensitive and specific, hence suitable for qualitative and quantitative analyses in pharmaceuticals, food science, clinical diagnostics.
